The purpose of this Notice of Special Interest (NOSI) is to promote the availability of Mentored Clinician Scientist Research Career Development Awards (K08) applications in research areas covered by the NIH BRAIN Initiative. This Notice of Funding Opportunity (NOFO) invites applications for basic, translational, and clinical research projects that propose to study the role of environmental stress on the health inequities in Alzheimer’s Disease Related Dementias (ADRD), including Frontotemporal Dementia (FTD), Lewy Body Dementia (LBD), Vascular Contributions to Cognitive Impairment and Dementia (VCID), and Mixed-Etiology Dementia (MED). Environmental stressors include but are not limited to heat, crowding, air pollution, noise, violence, and psychosocial stress. Many of these environmental stressors are more prevalent in rural and urban disadvantaged communities which also have a higher risk for ADRD. Thus, there is a need to identify environmental stressors that impact health inequities in ADRD and to determine the mechanistic underpinnings of this stress. This NOFO will support mechanistic, translational, and human subject studies to understand the relationship between environmental stressors and health inequities in ADRD. It is expected that these studies will identify environmental stressors related to health inequities in ADRD and their clinical relevance. Ultimately, these studies should accelerate research towards better therapeutic interventions and quality of life.

This Notice of Funding Opportunity (NOFO) supports the optimization of promising genome editing-based therapeutic leads for Alzheimer's Disease-Related Dementias (ADRD), towards IND-enabling studies. Specifically, it supports the characterization and optimization of therapeutic lead(s) that show promise as potential genome editing therapeutics, as evidenced by convincing proof-of-concept studies in appropriate models. At the end of the funding period, successful projects will have optimized a genome editing therapeutic candidate with demonstrated bioactivity, manufacturability, biodistribution, in vivo efficacy, and/or target engagement (measurement of proximal downstream effects) and optimal dosing, combined with other properties consistent with the desired clinical application. This NOFO is not specific for any one or group within the ADRD spectrum of disorders. The NeuroCOVID Project has collected and curated a wide range of data from the United States and international sites.
